
Xanthan gum serves as a popular thickener and stabilizer in many packaged foods today. Manufacturers add it to salad dressings, ice cream, gluten-free pasta, and protein powders for better texture. People with swallowing difficulties also rely on it to make liquids safer to consume. Recent research has raised questions about its long-term effects on the digestive system.
Scientists at the Federal University of São Paulo conducted a controlled experiment on adult male Wistar rats. They published their findings in the peer-reviewed journal PLOS One in April 2026. The team aimed to test whether chronic intake of xanthan gum could trigger inflammation in the colon. Their work builds on earlier clinical observations linking the additive to problems in premature infants.
Researchers divided thirty-two rats into four equal groups for the ten-week study. One group ate a standard diet without any xanthan gum as the control. The other three groups received diets containing one-quarter, one-half, or a full dose of the additive. These amounts were calculated to approximate different levels of human exposure, including everyday food use and medical thickening needs.
All three doses of xanthan gum led to signs of colon inflammation in the animals. Tissue samples showed higher numbers of lymphocytes, which are immune cells that gather during inflammatory responses. Levels of the inflammatory marker TNF-alpha also rose compared with the control group. Histological scores confirmed that the inflammation was mild to moderate overall.
The study also examined proteins that help maintain the intestinal barrier. Claudin-2, which regulates the passage of water and sodium between cells, showed increased expression in the treated rats. Another protein called ZO-1, involved in sealing the gut lining, also changed. These shifts suggested a possible loss of barrier integrity. Such changes can allow substances to pass more easily and may contribute to ongoing inflammation.
Gut bacteria profiles shifted as well after the xanthan gum diets. Overall diversity of the microbiota remained largely similar across groups. Major bacterial groups such as Bacteroidetes and Firmicutes did not change dramatically. However, the abundance of Elusimicrobiota increased, a change previously associated with inflammatory conditions in other research.
The findings offer experimental support for earlier concerns about necrotizing enterocolitis in premature infants. That serious condition had been linked observationally to xanthan gum used as a thickener in some formulas. The U.S. Food and Drug Administration later restricted its use in preemies after reported cases. This rat study demonstrated a causal relationship for inflammation under controlled conditions.
Lead researcher Alessandra Rischiteli and coordinator Claudia Oller emphasized caution rather than alarm. They noted that occasional small amounts of the additive may not pose problems for healthy adults. Their main concern centers on daily high intake and potential cumulative effects. Both stressed the importance of future human studies to confirm the results.
Several limitations apply to the current evidence. The experiment involved only rats, so direct translation to people remains uncertain. The sample size was relatively small with just eight animals per group. Dose conversions from humans to rodents also introduce some approximation. Inflammation observed was mild rather than severe in most cases.
Overall, this research highlights the value of examining common food additives more closely. Xanthan gum remains widely used and generally recognized as safe at typical levels by regulators. Individuals who consume large daily amounts, such as those managing dysphagia, may wish to discuss options with a healthcare provider. Continued investigation will help clarify any real-world risks for human gut health.
